Stem cell biology and technology
This course focuses on animal cell technologies and their applications. First, animal cell culture will be introduced as a basic technique and as an applied technique to perform genetic manipulation. The main themes will be : primary cultures and continuous cell lines, immortalization of cell lines, use of cell culture databases. In the next step, the course will focus on topics as recombinant protein and virus production, hybridomas for the production of antibodies, tissue-culture engineering, transgenesis, targeted gene inactivation, use of embryonic stem cells for generating chimeric animals and genetically-modified organisms. Finally the course will focus on the use of animal stem cells for research and in applied biomedical field. The following aspects will be studied : embryonic versus adult stem cells, pluri- and multipotency, primordial germ cells, trans-differentiation, nuclear re-programming, stem cell niches. The social and bioethical questions will be discussed in the last part of the course.
